The Study of Procyanidins' Effects on Prostate Cancer PC-3 Cells ...

[Abstract]

Prostate cancer, one of the most manifested male cancers, is in the second place among cancers leading male deaths in the developed countries. Procyanidins play an important role in preventing and curing oral cancer, skin tumor, breast cancer, pancreatic tumor, liver cancer, lung carcinoma, gastric adenocarcinoma and colon tumor, thanks to their antioxidant and free radical scavenging abilities as well as remarkably biological, pharmacological and therapeutic activities. The pharmacological effects of procyanidins on prostate cancer had been verified at the level of cell and sub-cell by our experiments. We testified for the first time that procyanidins were cytotoxicity and could induce the apoptosis and necrosis of PC-3 cells (human androgen-independent prostate cancer cell) which might be applied in the treatment of prostate cancer. The main experiments we performed were as follows:1. Effects of procyanidins on PC-3 cell proliferation MTT assay was applied to test the inhibitory effects of procyanidins on PC-3 cells proliferation. Different concentrations of procyanidins (100, 200 and 300 ug/ml) were incubated with PC-3 cells for 24, 48 and 72 hours, respectively. And procyanidins remarkably inhibited the proliferation of PC-3 cells in a dose- and time- dependent manner, and the IC50 was 155.7 ug/ml after 48 hours exposure.2. Effects of procyanidins on PC-3 cell morphology PC-3 cells? morphological changes caused by procyanidins at different concentrations were observed by invert microscopy, HE staining and transmission electron microscopy (TEM). Observed with invert microscopy, PC-3 cells appeared round and small with wrinkle as well as broken after incubated with different concentrations of procyanidins, which indicated that procyanidins were cytotoxicity to PC-3 cells. For TEM, there were classical characteristics of cell death including cell crimple, chromosome aggregation and break, cell membrane and mitochondrial membrane break, and there were bubbles in cells. All of these showed that procyanidins were able to inhibit the growth of PC-3 cells, change their morphology as well as induce necrosis. 3.
